Details
Specifically detects BRE1-A (RNF20) Clone: 8A4.2 in Human samples, and it is validated for Western BlottingE3 ubiquitin-protein ligase BRE1A (UniProt Q5VTR2; also known as BRE1 E3 ubiquitin ligase homolog, BRE1-A, hBRE1, Homolog of S. cerevisiae BRE1, RING finger protein 20) is encoded by the RNF20 (also known as BRE1, BRE1A) gene (Gene ID 56254) in human. BRE1A (RNF20) is a RING finger domain-containing protein originally identified as the E3 uqibiquitin ligase that catalyzes histone H2B Lys120 monoubiquitination in yeast. The histone chaperone FACT is reported to mediate RNF20 recruitment for chromatin remodeling to initiate homologous recombination repair (HRR). RNF20 is also shown to negatively regulate hepatic fatty acid metabolism by targeting SREBP1c for ubiquitination and subsequent proteasomal degradation. In addition, RNF20 is shown to mediated the polyubiquitination of ErbB3-binding protein 1 (Ebp1) and Transcription factor activator protein 2α (AP-2α). AP-2α functions as a negative regulator of adipogenesis by suppressing the transcription of CCAAT/enhancer binding protein (C/EBPα). Overexpression of RNF20 in 3T3-L1 cells results in a down-regulation of endogenous AP-2α and a concomitant transcriptional upregulation of C/EBPα.
