Specifically detects AHCYL1 (IRBIT) Clone: in Human samples, and it is validated for Western BlottingPutative adenosylhomocysteinase 2 (EC 3.3.1.1; UniProt O43865; also known as AdoHcyase 2, DC-expressed AHCY-like molecule, IP(3)Rs binding protein released with IP(3), IRBIT, S-adenosyl-L-homocysteine hydrolase 2, S-adenosylhomocysteine hydrolase-like protein 1) is encoded by the AHCYL1 (also known as DCAL, PPP1R78, PRO0233, XPVKONA) gene (Gene ID 10768) in human. Originally identified by its binding and antagonist activity against teh intracellular calcium channel, IP3R, IRBIT is now known to interact with multiple targets and regulate diverse physiological events in addition to blocking IP3 access to IP3R. IRBIT complexes with ribonucleotide reductase (RNR) in a dATP-dependent manner and stabilizes RNR in its inactive, dATP-bound state. IRBIT also targets CaMKIIα kinase for inhibition by blocking calmodulin binding. Other known IRBIT targets include the ion transporter NBCe1-B, the Na /H exchanger NHE3, the Cl- channel CFTR, the Cl-/HCO3- exchanger Slc26a6, the Fip1 component of the cleavage and polyadenylation specificity factor (CPSF) complex. IRBIT contains an S-adenosyl-l-homocysteine hydrolase (AHCY) homology domain in and an N-terminal extension containing a serine-rich region with multiple phosphorylation sites. Long-IRBIT, but not IRBIT, has an additional N-terminal appendage, termed the LISN (Long-IRBIT specific N-terminal) domain. IRBIT is highly conserved among species, and the human and mouse orthologs have 100% protein identity.
